17歳の少年が父親殺しの罪で起訴され、12名の陪審員たちがその事件の評決の為に集まってくる。目撃証言・証拠などから誰もが少年の有罪を信じて疑わない中、1人の陪審員が"無罪"を主張する。1人でも意見が違う場合は討論をして全員一致としなければならない、という陪審員制度に則って、12人の男たちは議論を開始。階級も職業も価値観も違う彼らの中には、さっさと討論を済ませて家に帰りたいと思っている者さえいた。しかし話し合いを続けるうちに、事件に関する新たな事実や疑惑が浮かび上がってきて、1人、また1人と無罪に賛成する者が増えていく。そして数時間に渡った討論は終わり、彼らが出した結論とは…。
