黒い帽子に黒のサングラス、黒いネクタイに黒のスーツという、全身黒づくめのジェイク・ブルースとエルウッド・ブルースは、ちぎりを交わした兄弟分。そのブルース兄弟が昔世話になった孤児院が、窮地に陥った！　彼らは孤児院を救おうと、かつての仲間を集めて“ブルース・ブラザース・バンド”を再結成し、そのコンサートの利益を孤児院に寄付しようとするが……。
