水木しげる原作の妖怪マンガを豪華キャストと最新技術で実写化し大ヒットを記録した『ゲゲゲの鬼太郎』の記憶も新しい中、更にパワーアップして贈る第2弾、1月9日にリリース決定！
小雨の降る夜に「かごめ歌」を聴いてしまった若い女性が失踪するという怪事件が続発していた。その事件に巻き込まれた女子高生・比良本楓（北乃きい）とともに、謎の解明に乗り出した鬼太郎（ウエンツ瑛士）たち。
妖怪図書館の司書妖怪・文車妖妃（中川翔子）の助けを得て、千年の時を経て蘇った悪霊の呪いがその原因であることを突き止める。呪いを封印するためには、古(いにしえ)の楽器を集め、"護人囃子(ごにんばやし)の儀(ぎ)"を行わなければならない。
刻一刻と楓に迫る呪いの期限。楽器を求め、古地図に記された「天」「地」「海」の場所へ分かれて旅立つ一行。鬼太郎と楓は「天」＝箱根の神山へ、猫娘（田中麗奈）とねずみ男（大泉洋）は「地」＝高尾山へ、砂かけ婆（室井滋）と子なき爺（間寛平）は「海」＝三浦半島の沖合へ出発する･･･。
